Migration of single immune cells in vitro : mechanisms of propulsion and guidance

Abstract : We use microfabricated tools and human primary lymphocytes to decipher amoeboid cell migration in vitro. Concerning propulsion, mammalian cells usually need adhesion or friction with a substrate to move. We show here that lymphocytes can swim and we propose a new mechanism of molecular paddling. Concerning guidance, we present new phenotypes of mechanical guidance by flow and of chemical guidance by adhesion molecules, and we discuss their active or passive nature.
